Anti-Hu CD20, 2H7, FI, 100tests

This FITC-conjugated mouse monoclonal antibody from STEMCELL Technologies, clone 2H7, is directed against the human CD20 antigen — an approximately 35 kDa non-glycosylated type I transmembrane protein of the MS4A family. The antibody is of the IgG2b, kappa isotype and was raised against human tonsillar B cells. It was purified by affinity chromatography and conjugated with FITC under optimal conditions, and the solution is free of unconjugated FITC.

CD20 is expressed on pre-B cells, resting and activated B cells, some follicular dendritic cells, and a subset of T cells, with expression lost when B cells differentiate into plasma cells. Clone 2H7 has been mapped to the amino acid sequence YNCEPANPSEKNSPST in the large extracellular loop of CD20. The reagent is verified for flow cytometry and has been used for purity assessments of cells isolated with EasySep™ kits; published reports also describe its use in immunocytochemistry, immunofluorescence, immunohistochemistry, ELISA, and functional assays.

Reported species reactivity includes human, rhesus, cynomolgus, baboon, chimpanzee, capuchin monkey, Nancy Ma's night monkey, pigtailed macaque, and squirrel monkey. For flow cytometry the manufacturer suggests 5 μL per 1 × 10⁶ cells in 100 μL volume or per 100 μL of whole blood, with titration recommended for optimal performance in each application. The product is supplied in a phosphate-buffered solution at pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and should be stored undiluted at 2 - 8 °C, protected from prolonged exposure to light and not frozen. According to the manufacturer's safety data sheet, the mixture is not classified as hazardous under the 2012 OSHA Hazard Communication Standard and is supplied as a clear liquid.
