Anti-Hu CD2, RPA-2.10, PR, 500tests

This PerCP-conjugated mouse monoclonal antibody from STEMCELL Technologies, clone RPA-2.10, targets the human CD2 antigen (Gene ID 914), also referred to as LFA-2, SRBC-R, or T11. It is of the IgG1, kappa isotype and was generated using human CD2 recombinant protein as immunogen. Flow cytometry (FC) is both the verified and reported application, with a suggested use of ≤ 0.125 µg per 1 × 10⁶ cells in 100 µL, and titration recommended for optimal performance.

CD2 is an approximately 50 kDa type I transmembrane glycoprotein and a member of the immunoglobulin superfamily, bearing two Ig-like domains in its extracellular portion. It is broadly expressed on peripheral T and NK cells, dendritic cells, erythrocytes, most thymic T cells, subsets of thymic B cells, and on the endothelium, and its expression appears early during T cell differentiation. Aberrant expression has been observed in some lymphomas and myeloid leukemias. CD2 plays a critical role in T cell activation and signaling as well as in lymphocyte adhesion; its primary ligand is CD58 (LFA-3) on antigen-presenting cells, with additional ligands including CD15 (SSEA-1), CD48, and CD59. The RPA-2.10 clone is known to block the mixed lymphocyte reaction. Reported species reactivity covers human, rhesus, cynomolgus, baboon, chimpanzee, capuchin monkey, pigtailed macaque, squirrel monkey, and pig.

The reagent is formulated in phosphate-buffered saline, pH 7.2, containing 0.09% sodium azide, 0.1% gelatin, and < 0.2% (w/v) bovine serum albumin. It was purified by affinity chromatography and conjugated with PerCP under optimal conditions, and the solution is free of unconjugated PerCP. The product is stable at 2–8 °C when stored undiluted; it should not be frozen and must be protected from prolonged exposure to light. The clone has been verified for purity assessments of cells isolated with EasySep kits, including the EasySep HLA Chimerism Whole Blood CD3 Positive Selection Kit. Per the manufacturer's safety data sheet it is a laboratory reagent and is not classified as a hazardous substance or mixture according to GHS, presented as a clear liquid.
