Flow cytometry workflows for human T cell immunophenotyping frequently rely on clone RPA-2.10, a mouse monoclonal antibody that specifically binds the human CD2 (LFA-2) glycoprotein. Provided by STEMCELL Technologies as a fluorescein isothiocyanate (FITC) conjugate, this reagent labels CD2-positive thymocytes, mature T lymphocytes, and natural killer cells for detection on the FITC channel of standard cytometers equipped with a 488 nm blue laser.

CD2 is a 50 kDa transmembrane receptor that mediates cell-cell adhesion through interaction with CD58 on antigen-presenting cells, making it a useful marker for tracking T cell activation, immune reconstitution after hematopoietic stem cell transplantation, and characterization of engineered CAR-T cell products. Each vial supplies enough antibody for 25 individual staining reactions, with approximately 5 uL of reagent per 100 uL cell suspension serving as a typical working dilution for freshly isolated peripheral blood mononuclear cells or whole blood specimens.

The fluorescein conjugate should be stored refrigerated and protected from light, with freeze-thaw cycles avoided to preserve fluorescence intensity. For research use only, this antibody supports both single-color and multicolor panel design when combined with other fluorophore-conjugated clones.
