Anti-Hu CD2, RPA-2.10, AZ, 100tests

STEMCELL Technologies supplies this anti-human CD2 reagent, Clone RPA-2.10, as a mouse monoclonal IgG1, kappa antibody conjugated to APC (Allophycocyanin). It was raised against a human CD2 recombinant protein and purified by affinity chromatography, and the solution is free of unconjugated APC and unconjugated antibody.

CD2 is an ~50 kDa type I transmembrane glycoprotein of the immunoglobulin (Ig) superfamily, displaying two Ig-like domains within its extracellular portion. The target is broadly expressed on peripheral T and NK cells, dendritic cells, erythrocytes, most thymic T cells, subsets of thymic B cells, and on the endothelium. CD2 expression appears early in T cell differentiation, and aberrant expression has been reported in some lymphomas and myeloid leukemias. CD2 plays a critical role in T cell activation and signaling, and in lymphocyte adhesion. Its primary ligand is CD58 (LFA-3), with additional ligands comprising CD15 (SSEA-1), CD48, and CD59. The RPA-2.10 clone is also noted to block the mixed lymphocyte reaction.

Reactivity has been demonstrated with human, rhesus, cynomolgus, baboon, chimpanzee, capuchin monkey, pigtailed macaque, squirrel monkey, and pig samples, and the antibody is verified for flow cytometry (FC). It has been validated for purity assessments of cells isolated with EasySep kits, including the EasySep HLA Chimerism Whole Blood CD3 Positive Selection Kit. For flow cytometry, the suggested use of this antibody is ≤ 0.03 µg per 1 x 10^6 cells in 100 µL, and titration is recommended for optimal performance for each application. The reagent is formulated in phosphate-buffered saline, pH 7.2, containing 0.09% sodium azide, 0.1% gelatin, and < 0.2% (w/v) bovine serum albumin. It is stable at 2 – 8 °C when stored undiluted; the product should not be frozen and should be protected from prolonged exposure to light. The product is intended for research use only.
