Anti-Hu CD184 (CXCR4), 12G5, 100ug

A mouse monoclonal IgG2a, kappa antibody supplied by STEMCELL Technologies, Clone 12G5, reacts with CD184 (CXCR4 or fusin), an ~45 kDa seven-transmembrane G-protein-coupled receptor expressed on the surface of a broad range of cells including T and B cells, dendritic cells, monocytes, macrophages, granulocytes, platelets, endothelial and epithelial cells, astrocytes and neurons. The antibody was generated in BALB/c mice using CP-MAC virus-infected SUP-T1 cells of human origin as immunogen and reacts with CD184 from human, rhesus, cynomolgus, baboon, chimpanzee, African green monkey, and sooty mangabey. At least two isoforms of CD184 are generated by alternate mRNA splicing, and ligands for the receptor include the CXC chemokine SDF-1 and ubiquitin.

Binding of Clone 12G5 blocks CD184 function as a co-receptor for infection by some T cell-tropic isolates of HIV-1 and as an alternative receptor for CD4-independent infection by HIV-2, and partially inhibits SDF-1-induced chemotaxis and calcium influx. In differentiation studies, CD184 is used alongside markers such as CD117 to monitor the emergence of definitive endoderm, and this clone has been verified for purity assessments of cells cultured with the STEMdiff™ Definitive Endoderm Kit. Verified applications are cell separation and flow cytometry; reported applications include CyTOF®, functional assay, flow cytometry, immunocytochemistry, immunofluorescence, and immunohistochemistry.

The antibody is supplied unconjugated in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and is purified by affinity chromatography. For flow cytometry, the suggested use is ≤ 1 μg per 1 x 10^6 cells in 100 μL volume or per 100 μL whole blood, with titration recommended for optimal performance for each application. The product is stable at 2 - 8°C when stored undiluted and should not be frozen.
