Anti-Hu CD14, M5E2, 100µg

This mouse monoclonal IgG2a, kappa antibody from STEMCELL Technologies targets CD14 (also known as LPS receptor), an ~53–55 kDa GPI-anchored transmembrane glycoprotein expressed at high levels on peripheral blood monocytes and macrophages and at lower levels on granulocytes. An ~10-fold difference in expression between monocytes/macrophages and granulocytes makes CD14 a useful marker for distinguishing these cell populations. The M5E2 clone was raised against full-length human CD14 protein and is supplied unconjugated in a phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide, and has been purified by affinity chromatography.

The antibody reacts with human, rhesus, cynomolgus, chimpanzee, capuchin monkey, common marmoset, cotton-topped tamarin, pigtailed macaque, squirrel monkey, cow, dog, pig, and sheep CD14. Flow cytometry is the verified application, with blocking, CyTOF®, functional assay, FC, immunocytochemistry, immunofluorescence microscopy, and immunohistochemistry reported in the literature; the M5E2 clone is not recommended for immunohistochemistry on formalin-fixed, paraffin-embedded sections. For flow cytometry, the suggested use is ≤ 2 μg per 1 x 10^6 cells in 100 μL. The antibody has been verified for purity assessments of cells isolated with EasySep™ cell separation kits.

CD14 functions as a high-affinity receptor for complexes of lipopolysaccharide (LPS) and serum LPS-binding protein and modulates LPS-dependent signal transduction during the immune response to gram-negative pathogens by acting as a co-receptor for TLR 4 and MD-2, triggering NF-kappa-B activation, cytokine secretion, and the inflammatory response. Two soluble forms of CD14 (~48 and ~55 kDa) have also been described. The product is stable at 2–8°C when stored undiluted; do not freeze.
