Anti-Hu CD123, 6H6, FI, 25tests

From STEMCELL Technologies, this FITC-conjugated mouse monoclonal IgG1, kappa antibody (Clone 6H6) is raised against human CD123 (IL-3 receptor subunit α; Gene ID 3563), an ~70 kDa type I transmembrane glycoprotein of the type I cytokine receptor family (type 5 subfamily) and the immunoglobulin superfamily. The antibody was generated by immunizing mice (BALB/c) with COS cells transfected with a cDNA encoding the human IL-3Rα chain, and cross-reacts with rhesus and sooty mangabey samples. CD123 constitutes the ligand-binding α chain of the heterodimeric IL-3 receptor and binds IL-3 with low affinity on its own, but achieves high-affinity IL-3 binding when associated with CD131.

CD123 is expressed by hematopoietic progenitor cells, endothelial cells, basophils, eosinophils, mast cells, monocytes, macrophages, dendritic cells, megakaryocytes, a subset of B cells, and by neutrophils when cultured in the presence of GM-CSF. IL-3 engagement of CD123 stimulates proliferation, differentiation, and viability of hematopoietic cells, and CD123 is highly expressed in malignancies such as acute leukemia. Clone 6H6 does not inhibit binding of IL-3 to CD123 or to the IL-3 receptor. The reagent is supplied in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, is purified by affinity chromatography, and is free of unconjugated FITC.

Verified for flow cytometry, the antibody has been used to assess the purity of cells isolated with EasySep™ kits, including the Human Plasmacytoid DC Enrichment Kit and the Human Basophil Enrichment Kit. The manufacturer's suggested use is ≤ 5 μL per 1 x 10^6 cells in 100 μL, with titration recommended for optimal performance. The product is stable at 2 - 8°C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light.
