Description

STEMCELL Technologies supplies a FITC-conjugated mouse monoclonal antibody specific for human CD11b (integrin αM), raised against the ICRF44 clone. The reagent is provided as a clear liquid and is produced under a quality management system certified to ISO 13485. Per the supplier, the product is for research use only and not intended for human or animal diagnostic or therapeutic applications unless otherwise stated.

CD11b is an ~170 kDa type 1 transmembrane glycoprotein that associates non-covalently with CD18 to form the heterodimeric Mac-1 receptor. Through interactions with ligands such as ICAM-1 (CD54), ICAM-2 (CD102), ICAM-4 (CD242), iC3b, heparin, and fibrinogen, Mac-1 influences adherence of neutrophils and monocytes to stimulated endothelium, and phagocytosis of complement-coated particles. The antigen is expressed on granulocytes, monocytes, NK cells, dendritic cells, tissue macrophages, and subsets of T and B cells, and is used as a marker to distinguish naïve and memory CD8+ T cells. CD11b is a relatively late marker of myeloid differentiation and is undetectable on most myelomonocytic hematopoietic progenitor cells and more primitive cells. Reported reactive species include human, chimpanzee, baboon, cynomolgus macaque, rhesus macaque, common marmoset, and pig (Gene ID: 3684).

The ICRF44 clone has been verified for flow cytometry on human peripheral blood mononuclear cells and whole blood nucleated cells, and for labeling human mesenchymal cells cultured in MesenCult™ Proliferation Kit (Human) and MesenCult™-XF Medium. The antibody is of the IgG1, kappa isotype and reportedly inhibits leukocyte aggregation in response to the chemoattractant fMLP. Documented applications include cell separation, CyTOF, flow cytometry, functional assays, immunocytochemistry, immunofluorescence, immunohistochemistry, immunoprecipitation, and Western blot. According to the manufacturer's safety data sheet, the mixture is not classified as hazardous under the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200) or the Globally Harmonized System, with 0 % of the mixture consisting of ingredient(s) of unknown toxicity. The product is chemically stable under normal conditions, and containers should be kept tightly closed in a dry, cool, and well-ventilated place.
