Anti-Hu CD105, 43A3, PE, 25tests

Manufactured by STEMCELL Technologies, this PE-conjugated monoclonal antibody (clone 43A3) targets CD105 (endoglin), an approximately 180 kDa cell surface glycoprotein that forms a disulfide-bonded homodimer of ~90 kDa type I transmembrane subunits. The reagent is a mouse IgG1, kappa antibody raised against L-cells transfected with human CD105, with verified reactivity against human and mouse samples. CD105 is a component of the TGF-β receptor complex, expressed by vascular endothelial smooth muscle cells, syncytiotrophoblasts of the placenta, activated macrophages, and at relatively low levels by stromal fibroblasts, with expression also observed in some types of tumors and levels up-regulated on the endothelium during angiogenesis.

According to the manufacturer's product literature, in concert with signaling receptors the target binds TGF-β1 and TGF-β3 with high affinity but does not bind TGF-β2, with other reported ligands including Activin A, BMP-2, and BMP-7. The protein has important roles in angiogenesis, cardiovascular development, and vascular remodeling, and serves a regulatory role in cytoskeletal reorganization by modulating the sites of focal adhesion and cellular migration; certain mutations in CD105 result in the autosomal dominant disorder hereditary hemorrhagic telangiectasia. Flow cytometry (FC) is the verified application, and the clone has been verified for labeling human mesenchymal cells grown in MesenCult™ Proliferation Kit (Human).

The reagent is supplied as a phosphate-buffered solution at pH 7.2, containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and the antibody has been purified by affinity chromatography. The associated safety data sheet describes the product as a clear liquid that is not classified as hazardous under the 2012 OSHA Hazard Communication Standard, with handling guidance recommending that containers be kept tightly closed in a dry, cool, and well-ventilated place.
