Description

A mouse monoclonal IgG1, kappa antibody manufactured by STEMCELL Technologies targets CD105 (endoglin), a component of the TGF-β receptor complex. The 43A3 clone is conjugated with FITC and reacts with both human and mouse CD105; it has been verified by the manufacturer for flow cytometry applications.

CD105 is an approximately 180 kDa cell-surface glycoprotein that forms a disulfide-bonded homodimer of roughly 90 kDa type I transmembrane subunits. Expression has been reported on vascular endothelial smooth-muscle cells, placental syncytiotrophoblasts, and activated macrophages, and at relatively low levels on stromal fibroblasts. It is also detected on some tumors and is upregulated on the endothelium during angiogenesis. In concert with signaling receptors, CD105 binds TGF-β1 and TGF-β3 with high affinity but does not bind TGF-β2; other reported ligands include Activin A, BMP-2, and BMP-7. Mutations in CD105 have been linked to the autosomal dominant disorder hereditary hemorrhagic telangiectasia.

The antibody was raised against L cells transfected with human CD105 and purified by affinity chromatography; the FITC conjugate is described as free of unconjugated FITC. It is supplied in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide and 0.2% (w/v) bovine serum albumin, and is stable at 2–8 °C when stored undiluted. The product should not be frozen and should be protected from prolonged exposure to light. For flow cytometry, the manufacturer suggests using ≤ 5 µL per 1 × 10⁶ cells in 100 µL, and recommends titration for each application.
