Description

A biotin-conjugated monoclonal antibody from STEMCELL Technologies, Clone AC384, recognizes angiotensin-converting enzyme 2 (ACE2), a type I integral membrane protein with a short C-terminal cytoplasmic domain of 22 amino acids, a transmembrane domain, and a large N-terminal ectodomain of 740 amino acids. The antibody was raised against the extracellular domain of human ACE2 recombinant protein (aa 18 – 740) and is of the mouse IgG1, kappa isotype. It is reported for use in ELISA, flow cytometry, functional blocking, and Western blotting, with suggested starting dilutions of 1:2,000 – 1:10,000 for ELISA and 1:2,000 – 1:10,000 for Western blotting; titration is recommended for each application. The reagent is supplied in phosphate-buffered solution, pH 7.4, was purified by affinity chromatography, and is stable at -20°C when stored undiluted.

ACE2 contains a catalytic zinc-binding motif within its N-terminal domain that shares approximately 42% similarity with the catalytic binding domain of ACE; however, ACE2 functions as a carboxypeptidase, whereas ACE acts as a peptidyl dipeptidase or endopeptidase. In humans, ACE2 is a key regulator of the renin-angiotensin system, where it catalyzes the cleavage of the vasoconstrictor peptide angiotensin II to angiotensin 1-7. ACE2 expression can be found on endothelial cells throughout the body, including the kidney, heart, and lungs. In the airways leading to the lungs, ACE2 provides the main entry point for SARS-CoV-2; once the viral Spike protein binds to ACE2, fusion with the host cell membrane can occur, resulting in delivery of the viral RNA genome.
