Description

Assay Genie provides a cell-based ELISA kit intended for the quantitative analysis of ZNF174 levels in adherent cell cultures, cell lysates, and tissue samples. The assay uses a colorimetric detection method read at 450 nm and is supplied in a 96-Well Microplate format. The kit is reactive against human samples, with a stated dynamic range of > 5000 Cells, and is stored at 4°C with a stability of 6 months. ZNF174 is a zinc-finger protein of the krueppel C2H2-type family that acts as a transcriptional repressor; two splice variant isoforms have been described, and its dysregulation has been implicated in cancer and autoimmune disorders, making it a valuable biomarker for studying disease mechanisms and developing targeted therapies.

Qualitative determination of ZNF174 concentration is achieved by an indirect ELISA format in which ZNF174 is captured by ZNF174-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to act as an internal positive control in normalizing the target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts.

Database identifiers for the ZNF174 target include NCBI Gene ID 7727, UniProt ID Q15697, OMIM 603900, and Unigene Hs.155204, with a chromosomal location of 16p13.3 and a molecular weight of 26,884 Da. The protein is also known by the synonym names AW-1, Zinc finger and SCAN domain-containing protein 8, and ZSCAN8, and the kit falls within the Epigenetics and Nuclear Signaling research area.
