Description

Assay Genie provides a Sandwich ELISA platform for the quantitative detection of MIP-3b (also known as CCL19) in human samples. The assay uses a double-antibody sandwich configuration with colorimetric readout, intended for high sensitivity and specificity across a range of research applications.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit delivers a measurable range of 15.625–1000 pg/mL with a sensitivity of 9.375 pg/mL, and reagents are intended to be stored at 2–8°C for up to 12 months. Reported linearity is 85–105% (1:2), 91–105% (1:4) and 86–101% (1:8) for serum; 82–100%, 82–97% and 83–100% for EDTA plasma; and 84–99%, 87–98% and 84–100% for heparin plasma. Average recovery is 91% for serum (range 86–96%), 92% for EDTA plasma (range 86–102%) and 95% for heparin plasma (range 88–102%).

The workflow involves equilibrating reagents and TMB substrate to room temperature, loading standards and samples for a 90-minute primary incubation at 37°C, sequential incubations with biotin-labelled detection antibody (60 minutes) and HRP-Streptavidin (30 minutes), TMB substrate incubation in the dark for 10–20 minutes, and absorbance reading at 450 nm after adding stop solution. MIP-3b/CCL19 is a chemokine that attracts immune cells to sites of inflammation and infection, with roles in T cell activation, lymphocyte trafficking and dendritic cell migration. Dysregulation of MIP-3b levels has been associated with inflammatory diseases, autoimmune disorders and cancer, underscoring its value as a biomarker for research into these conditions.
