Description

A high-throughput, lysate-free Cell-Based ELISA from Assay Genie enables sensitive detection and quantification of phosphorylated ZAP-70 at tyrosine 493 in cell lysates and tissue homogenates. The assay provides a convenient way to study ZAP-70 protein phosphorylation and expression profiles in cultured cells.

ZAP-70 is a key signaling molecule in T cells, playing a critical role in T cell receptor signaling and activation. Phosphorylation of ZAP-70 at Tyr493 is crucial for its function in regulating T cell responses and immune system function. This kit offers high sensitivity and specificity, making it suitable for research studies involving signal transduction pathways and immune response.

The assay uses an indirect ELISA format where the target is captured by phospho-specific primary antibodies, and HRP-conjugated secondary antibodies enable colorimetric detection at 450 nm. The kit is reactive with Human, Mouse, and Rat samples, with a dynamic range of > 5000 cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and Crystal Violet whole-cell staining may be used to determine cell density. Researchers can use this kit to measure the relative amounts of phosphorylated ZAP-70 in cultured cells as well as screen for the effects of various treatments, inhibitors, or activators on ZAP-70 phosphorylation.
