Description

A lysate-free, high-throughput cell-based ELISA from Assay Genie is designed to detect XPF (Xeroderma Pigmentosum, complementation group F) protein levels in cells. The assay uses an indirect ELISA format with HRP-conjugated secondary antibodies to catalyze a colorimetric reaction at 450 nm.

The kit can be used to measure relative amounts of XPF in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on XPF expression. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density.

XPF is encoded by the ERCC4 gene (Gene ID: 2072, UniProt ID: Q92889) and is a catalytic component of a structure-specific DNA repair endonuclease responsible for the 5-prime incision during DNA repair. The protein has a molecular weight of 103 kDa and is involved in nucleotide excision repair. The kit shows reactivity with human and mouse samples, has a dynamic range of > 5000 Cells, and is suitable for adherent cells. Storage is at 4°C for 6 months. Applications include research in epigenetics and nuclear signaling, with relevance to cancer, DNA repair, and genomic stability.
