Description

Assay Genie provides a Cell-Based ELISA Kit for the measurement of XPA (Xeroderma Pigmentosum Group A) levels in cell lysates and tissue samples. Designed for studies of DNA repair mechanisms, genetic disorders and cancer biology, the kit targets a zinc finger protein involved in DNA excision repair, particularly in the recognition and removal of UV radiation-induced DNA damage. Dysregulation of XPA has been linked to Xeroderma Pigmentosum, Complementation Group A, and to certain cancers.

Detection is by colorimetric readout at 450 nm in a 96-well microplate format, with a stated dynamic range of > 5000 cells and reactivity in Human and Mouse samples. The kit is described as lysate-free, high-throughput and sensitive, and is intended for measuring relative XPA amounts in cultured cells as well as for screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on XPA. Qualitative determination is achieved through an indirect ELISA format in which XPA-specific primary antibodies capture the target and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition.

A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalising target absorbance values. Following the colorimetric measurement of HRP activity, a Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalised to cell amounts and correcting for plating differences. The associated XPA protein is listed under UniProt ID P23025 and Gene ID 7507, with a reported molecular weight of 31,368 Da and a chromosomal location of 9q22.3 in humans. Storage is at 4°C with a stability of 6 Months, and the kit is classified within the Epigenetics and Nuclear Signaling research area.
