XIAP Colorimetric Cell-Based ELISA Kit

Assay Genie's cell-based ELISA kit enables colorimetric detection of XIAP (X-linked Inhibitor of Apoptosis Protein) directly in cell lysates and culture supernatants using an indirect ELISA format. Target-specific primary antibodies capture XIAP in the well, and HRP-conjugated secondary antibodies catalyse a colour-forming reaction read at 450 nm, providing a sensitive and reproducible measurement of relative XIAP protein levels in cultured cells.

Reactivity has been validated for Human, Mouse, and Rat samples, and the assay supports a dynamic range of > 5000 Cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining step is used to determine cell density so that absorbance values can be adjusted for plating variation. The protocol is suited to adherent samples and is designed for researchers measuring relative XIAP expression, screening treatment effects, or evaluating the impact of inhibitors such as siRNA or chemical modulators on XIAP levels.

XIAP is a key regulator of apoptosis and cell survival, and dysregulation has been implicated in cancer, autoimmune disorders, and neurodegenerative disease, making the assay a useful tool for disease-mechanism and therapeutic-target studies. The kit should be stored at 4 °C and is stable for 6 months under those conditions.
