Description

Assay Genie's cell-based ELISA kit is designed to detect and quantify levels of the WWOX (WW domain-containing oxidoreductase) protein in cell lysates, tissue homogenates, and other biological samples. WWOX is a tumor suppressor gene that plays a critical role in regulating cell growth, apoptosis, and DNA repair processes, and its dysregulation has been linked to various cancers, making it a relevant biomarker for cancer research and potential therapeutic studies.

The assay uses a colorimetric detection method measured at 450 nm in a 96-well microplate format and is reactive with human and mouse samples. It is a lysate-free, high-throughput workflow based on an indirect ELISA format, in which WWOX-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts.

The kit has a dynamic range of more than 5,000 cells and is intended for qualitative measurement of relative WWOX levels in cultured cells, including screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators. The product is suitable for use with adherent cells, and is stored at 4°C with a stated stability of 6 months.
