Description

A Cell-Based ELISA kit from Assay Genie designed for the colorimetric measurement of WTAP (Wilms Tumor 1-Associated Protein) in cell lysates and culture supernatants. The assay is reactive with human and mouse samples and uses an indirect ELISA format, in which WTAP is captured by specific primary antibodies and detected via HRP-conjugated secondary antibodies, producing a colorimetric read-out at 450 nm. Described as lysate-free, high-throughput, and sensitive, the kit can be used to assess relative levels of WTAP in cultured cells and to screen the effects of treatments, inhibitors (including siRNA or chemicals), or activators on WTAP expression.

The kit supports qualitative determination of WTAP concentration through two normalization approaches: a monoclonal anti-GAPDH antibody supplied as an internal positive control, and a Crystal Violet whole-cell staining step used to determine cell density. The recommended sample type is adherent cells, and the assay has a stated dynamic range of > 5000 Cells. Storage is recommended at 4°C with a shelf life of 6 months.

WTAP is a ubiquitously expressed nuclear protein involved in RNA modification and regulation, with roles in gene expression, cell proliferation, and mRNA splicing. It regulates the G2/M cell-cycle transition by binding to the 3' UTR of CCNA2, and has been implicated in cancer, neurological disorders, and developmental abnormalities, making it a target of interest in biomarker and therapeutic research.
