Description

From Assay Genie, this cell-based ELISA kit is designed to measure phosphorylated WAVE1 protein at the Tyr125 site in cell samples. It uses a colorimetric detection method with absorbance read at 450 nm and is reactive with human, mouse, and rat samples. The kit is intended for adherent cells and works through an indirect ELISA format, in which WAVE1 (Phospho-Tyr125) is captured by specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition.

The assay can be used to measure relative amounts of phosphorylated WAVE1 in cultured cells, as well as to screen for the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on WAVE1 phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step may be used after colorimetric measurement to determine cell density and adjust for plating differences. The dynamic range of the assay is > 5000 Cells.

WAVE1 (gene symbol WASF1) is a downstream effector involved in transmitting signals from tyrosine kinase receptors and small GTPases to the actin cytoskeleton. It promotes the formation of actin filaments as part of the WAVE complex, which regulates lamellipodia formation and actin filament reorganization via interaction with the Arp2/3 complex, and belongs to the SCAR/WAVE family. Phosphorylation of WAVE1 at the Tyr125 site has been implicated in cellular signaling pathways relevant to cancer metastasis, neuronal development, and other research areas.
