Description

Manufactured by Assay Genie, this WAVE1 Colorimetric Cell-Based ELISA Kit is a cell-based immunoassay for the qualitative detection of WAVE1 protein expression directly in cultured cells. It is reactive with human, mouse, and rat samples and uses a colorimetric detection method read at 450 nm, with a stated dynamic range of > 5000 Cells. The lysate-free, high-throughput workflow is run on a 96-Well Microplate format, and the kit is stored at 4°C with a stability of 6 Months. The kit is intended for adherent cell samples.

Detection is achieved through an indirect ELISA format in which WAVE1 is captured by WAVE1-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the conjugated HRP to catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control to normalise target absorbance values, and a Crystal Violet whole-cell staining method is provided to determine cell density and adjust for plating differences. The kit can also be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), and activators on WAVE1.

WAVE1 (gene symbol WASF1; NCBI Gene ID 8936; UniProt ID Q92558; molecular weight 61,652 Da) is a downstream effector involved in transmitting signals from tyrosine kinase receptors and small GTPases to the actin cytoskeleton, where it promotes actin filament formation. It operates as part of the WAVE complex, which regulates lamellipodia formation and actin filament reorganisation via interaction with the Arp2/3 complex, and belongs to the SCAR/WAVE family.
