Description

From Assay Genie, this colorimetric cell-based ELISA kit is designed for the accurate measurement of Vitamin D Receptor activation in human cells. It enables qualitative determination of Vitamin D Receptor protein expression in cultured cells through an indirect ELISA format, in which the target is captured by Vitamin D Receptor-specific primary antibodies and detected using HRP-conjugated secondary antibodies. Following substrate addition, the conjugated HRP catalyses a colorimetric reaction with detection at 450 nm.

The assay is described as convenient, lysate-free, high throughput and sensitive, with a quoted dynamic range of > 5000 Cells. It is intended for use with adherent cells and supports applications including drug discovery, cancer research, and studies on the role of Vitamin D in immune function and metabolism. Researchers can use the kit to measure relative amounts of Vitamin D Receptor in cultured cells and to screen for the effects of treatments, inhibitors such as siRNA or chemicals, or activators. Because the readout is qualitative, two normalisation approaches are provided: a monoclonal antibody specific for human GAPDH is included as an internal positive control, and Crystal Violet whole-cell staining can be used to determine cell density and normalise absorbance values to cell amounts.

The target is the VDR gene product (Gene ID: 7421; UniProt ID: P11473), a nuclear hormone receptor that mediates the action of vitamin D3. The kit is stored at 4 °C with a stated stability of 6 Months.
