Description

A Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the detection of vimentin levels in cell lysates and tissue samples, offering high sensitivity and specificity for accurate and reproducible results. Vimentin is a key intermediate filament protein that plays a critical role in maintaining cellular structure and motility, with aberrant expression linked to diseases such as cancer, fibrosis, and neurodegenerative disorders, making it an important biomarker for studying these conditions. The kit is intended for use in immunology research with adherent cell samples.

This convenient, lysate-free, high-throughput assay detects vimentin protein expression profiles in cells and can be used to measure the relative amounts of vimentin in cultured cells as well as to screen for the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on vimentin. Qualitative determination of vimentin concentration is achieved by an indirect ELISA format: vimentin is captured by vimentin-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the HRP enzyme to catalyze a colorimetric reaction upon substrate addition. The assay is reactive with human, mouse, and rat samples and is read at 450 nm.

For data normalization, a monoclonal antibody specific for human GAPDH is included as an internal positive control in normalizing the target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density after the colorimetric measurement. The dynamic range is greater than 5000 cells, and the kit is provided in a 96-well microplate format with a storage stability of 4°C for 6 months.
