Description

Designed for studying the activation status of the VEGFR2 signaling pathway in cell-based assays, this Assay Genie kit enables quantitative detection of phosphorylated VEGFR2 at Tyr951 in cell lysates. It is a lysate-free, high-throughput and sensitive assay for measuring VEGFR2 protein phosphorylation and expression profile in cells, with relevance to research in angiogenesis, vascular development, cancer biology, cardiovascular diseases and regenerative medicine. The sample type is adherent cells.

The kit uses an indirect ELISA format in which VEGFR2 (Phospho-Tyr951) is captured by specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The HRP enzyme conjugated to the secondary antibody catalyses a colorimetric reaction upon substrate addition, with detection measured colorimetrically at 450 nm and a dynamic range of > 5000 Cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values. Following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts.

The target is VEGFR2 (Phospho-Tyr951), with reactivity to human and mouse samples. VEGFR2, also known as KDR, is a receptor tyrosine kinase of the VEGFR family and a high affinity receptor for VEGF and VEGF-C, playing a key role in vascular development and the regulation of vascular permeability.
