Description

Designed by Assay Genie, this in-cell ELISA kit is configured to measure phosphorylation of Vascular Endothelial Growth Factor Receptor 2 (VEGFR2) at tyrosine 1214 in adherent cultured cells. The assay is a cell-based colorimetric ELISA read at 450 nm, using an indirect format in which VEGFR2 (Phospho-Tyr1214) is captured by phospho-specific primary antibodies while HRP-conjugated secondary antibodies catalyze the substrate reaction.

The assay has a dynamic range of more than 5000 cells and is validated for reactivity against human, mouse, and rat samples. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance, and a Crystal Violet whole-cell staining step can be performed to determine cell density.

VEGFR2 is a receptor tyrosine kinase of the VEGFR family and a high-affinity receptor for VEGF and VEGF-C, with ligand binding inducing autophosphorylation and activation. Phosphorylation at tyrosine 1214 is critical for downstream signaling that regulates endothelial cell function and vascular development, and dysregulation of VEGFR2 phosphorylation has been implicated in cancer, diabetic retinopathy, and cardiovascular disorders. The gene symbol is KDR and the protein is also known as kinase insert domain receptor.
