Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the accurate detection of phosphorylated VEGFR2 at tyrosine 1059 in cultured cells. The lysate-free, high-throughput assay uses a colorimetric detection method to enable qualitative measurement of VEGFR2 protein phosphorylation and expression profile, making it suitable for screening the effects of various treatments, inhibitors, or activators on VEGFR2 phosphorylation.

Detection is performed by an indirect ELISA format at 450 nm, with HRP-conjugated secondary antibodies binding the Fc region of VEGFR2 (Phospho-Tyr1059)-specific primary antibodies to catalyze a colorimetric reaction upon substrate addition. The kit is suited to adherent cell samples and shows reactivity with Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells. Because the assay is qualitative in nature, the kit includes a monoclonal anti-GAPDH antibody as an internal positive control for normalisation, and a Crystal Violet whole-cell staining method for determining cell density after measurement.

VEGFR2, also known as KDR (UniProt P35968, Gene ID 3791), is a receptor tyrosine kinase with high affinity for VEGF and VEGF-C. Ligand binding induces autophosphorylation and activation, recruiting downstream signalling proteins including Shc, GRB2, PI3K, Nck, SHP-1 and SHP-2, and playing a key role in vascular development and regulation of vascular permeability.
