Description

A cell-based ELISA kit from Assay Genie designed for the measurement of VEGFR2 in cell lysates and culture supernatants. The assay uses a colorimetric detection method at 450 nm and is reactive with Human, Mouse, and Rat samples, with a dynamic range of > 5000 cells. The lysate-free, high-throughput workflow enables sensitive detection of VEGFR2 protein expression profiles in cultured cells, including adherent cell samples.

The kit employs an indirect ELISA format, in which VEGFR2 is captured by target-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. Because the assay is qualitative, two normalization approaches are provided: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining method allows researchers to determine cell density and adjust absorbance values for plating differences. The kit can be used both to measure relative VEGFR2 levels in cultured cells and to screen for the effects of treatments, inhibitors such as siRNA or chemicals, and activators on VEGFR2 expression.

VEGFR2 (also known as KDR, Gene ID 3791, UniProt P35968) is a receptor tyrosine kinase of the VEGFR family and a high-affinity receptor for VEGF and VEGF-C. Ligand binding induces autophosphorylation and activation, and the receptor plays a key role in vascular development and regulation of vascular permeability; its dysregulation has been implicated in numerous diseases, including cancer and cardiovascular disorders. The kit is stored at 4°C with a stated stability of 6 months.
