Description

From Assay Genie, this cell-based ELISA platform is designed to detect VASP phosphorylated at serine 238 in cell lysates using a lysate-free, high-throughput workflow. The assay uses an indirect ELISA format in which a phospho-specific primary antibody captures the target, and an HRP-conjugated secondary antibody bound to the Fc region of the primary antibody catalyses a colorimetric reaction upon substrate addition. Absorbance is read at 450 nm. The kit is intended for adherent cells and can be used to measure relative levels of phosphorylated VASP in cultured cells, as well as to screen the effects of various treatments, inhibitors, or activators on VASP phosphorylation.

Reactivity has been validated for human, mouse, and rat samples, with a stated dynamic range of > 5000 Cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining step is provided to determine cell density, after which results can be normalised to cell amounts to adjust for plating differences. The kit is supplied with 96-well cell culture clear-bottom microplates, 10X TBS, wash, blocking and quenching buffers, primary antibody diluent, anti-phospho and total target antibodies, anti-GAPDH antibody, HRP-conjugated anti-rabbit and anti-mouse IgG secondary antibodies, SDS solution, stop solution, ready-to-use substrate, crystal violet solution, and adhesive plate seals.

VASP (vasodilator-stimulated phosphoprotein) is described by the manufacturer as an actin- and profilin-binding microfilament-associated protein whose phosphorylation is dynamically regulated by cellular adhesion to the extracellular matrix. Phosphorylation at Ser238 is highlighted as a key signalling event in regulating actin dynamics and cell migration, with dysfunction implicated in cancer, cardiovascular disorders, and neurological conditions. The kit is presented as easy to use with quick assay times, suited to researchers investigating the role of VASP phosphorylation in cellular processes and disease pathology.
