Description

Designed for accurate measurement of UBF1 in cultured cells, this Cell-Based ELISA Kit from Assay Genie employs a colorimetric detection method at 450 nm. It is described as a convenient, lysate-free, high-throughput and sensitive assay that detects the UBF1 protein expression profile in cells. The kit is suitable for measuring relative amounts of UBF1 in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on UBF1. Reactivity covers Human, Mouse and Rat samples, with a dynamic range of > 5000 Cells, and the assay is run on a 96-well microplate format using adherent cells. Storage is at 4 °C for 6 months.

Qualitative determination of UBF1 concentration is achieved through an indirect ELISA format, in which UBF1-specific primary antibodies capture the target and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density so that absorbance values can be normalised to cell amounts.

UBF1, also known as upstream binding factor 1 (gene symbol UBTF; Gene ID 7343; UniProt ID P17480), is a nucleolar transcription factor that recognises the ribosomal RNA gene promoter and activates transcription mediated by RNA polymerase I through cooperative interactions with the SL1/TIF-IB complex. It plays a critical role in regulating ribosomal DNA transcription and is encoded as a protein of 84,937 Da. The associated research area is Epigenetics and Nuclear Signaling.
