Description

From Assay Genie, this cell-based ELISA kit is designed for the detection of UBE1L levels in cell lysates and tissue samples. It provides a convenient, lysate-free, high-throughput and sensitive assay for measuring the relative amounts of UBE1L in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on UBE1L expression.

Detection proceeds via an indirect ELISA format: UBE1L is captured by UBE1L-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, enabling HRP to catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control in normalizing target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences. Readout is colorimetric at 450 nm, with a stated dynamic range of greater than 5000 cells.

The kit is reactive with human samples, intended for use with adherent cells, and is supplied in a 96-well microplate format with storage at 4°C for 6 months. UBE1L, also known as Ubiquitin-Activating Enzyme E1-like protein (gene symbol UBA7), is a key enzyme in the ubiquitination pathway involved in protein degradation and regulation. Dysregulation of UBE1L has been implicated in cancer, neurodegenerative disorders, and autoimmune diseases, making it a valuable target for research in molecular biology and drug discovery within the signal transduction research area.
