Description

From Assay Genie, this cell-based ELISA kit is designed for the detection of tyrosine hydroxylase phosphorylated at serine 8. It provides a lysate-free, high-throughput workflow for measuring the relative amounts of phosphorylated tyrosine hydroxylase in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on tyrosine hydroxylase phosphorylation. The assay uses an indirect ELISA format in which phospho-Ser8 tyrosine hydroxylase is captured by a specific primary antibody, with an HRP-conjugated secondary antibody catalysing a colorimetric reaction measured at 450 nm.

Reactivity is reported for rat samples, with adherent cells as the indicated sample type and a dynamic range of > 5000 cells. Two normalization strategies are built into the protocol: a monoclonal anti-GAPDH antibody serves as an internal positive control, and Crystal Violet whole-cell staining can be used to adjust absorbance values for cell density and plating differences. The target is tyrosine hydroxylase (Gene ID 25085; UniProt P04177; molecular weight 55,966 Da), a key enzyme in catecholamine biosynthesis whose phosphorylation at Ser8 regulates its activity and influences dopamine synthesis and neurotransmission. This makes the kit a useful tool for research into the molecular mechanisms underlying neurological and psychiatric disorders such as Parkinson's disease, schizophrenia, and addiction.
