Description

From Assay Genie, this colorimetric cell-based ELISA kit enables quantitative detection of Tubulin beta in cell lysates and cell culture supernatants using adherent cell samples. The assay is described as offering high sensitivity and specificity for accurate and reproducible results, suitable for research into a protein that is a key constituent of microtubules and is involved in cell division, intracellular transport, and cell structure. Dysregulation of Tubulin beta has been implicated in cancer, neurodegenerative disorders, and developmental abnormalities.

The workflow uses an indirect ELISA format in which target-specific primary antibodies capture Tubulin beta, and HRP-conjugated secondary antibodies catalyse a colorimetric reaction upon substrate addition. Reactivity covers Human, Mouse, and Rat samples, with a stated dynamic range of "> 5000 Cells" and detection read at 450 nm. The kit is lysate-free and high-throughput, and can be used to measure relative Tubulin beta amounts in cultured cells as well as to screen the effects of treatments, inhibitors such as siRNA or chemicals, or activators.

A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density. The kit is supplied in a 96-well microplate format with recommended storage at 4°C for 6 months. The target is encoded by the gene symbol TUBB3 (Gene ID: 10381, UniProt ID: Q13509, OMIM: 602661, Unigene: Hs.511743).
