Description

From Assay Genie, this cell-based ELISA kit enables the qualitative detection of Tuberin/TSC2 protein phosphorylation at Ser939 in cultured cells. The assay uses an indirect ELISA format in which target-specific primary antibodies capture Tuberin/TSC2 (Phospho-Ser939) and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, producing a colorimetric signal read at 450 nm. The kit is reactive with Human, Mouse, and Rat samples, has a dynamic range of > 5000 Cells, and is intended for use with adherent cells.

Researchers can use the kit to measure the relative levels of phosphorylated Tuberin/TSC2 in cultured cells and to screen the effects of treatments, siRNA, chemical inhibitors, or activators on Tuberin/TSC2 phosphorylation. Because the assay is qualitative, two normalization methods are supplied: a monoclonal anti-GAPDH antibody serves as an internal positive control for target absorbance values, and a Crystal Violet whole-cell staining step is used to determine cell density, so absorbance values can be normalized to cell amounts and corrected for plating differences.

Phosphorylation of TSC2 at Ser939 is an important regulatory event in the mTOR signaling pathway, which governs cell growth, proliferation, and metabolism. Dysregulation of this pathway has been associated with cancer, neurodevelopmental disorders, and metabolic syndromes, making this phospho-specific, immunology-focused cell-based ELISA kit a useful tool for studying mTOR-related signaling and disease pathogenesis.
