Description

This Cell-Based ELISA Kit from Assay Genie enables colorimetric detection of Tuberin/TSC2 protein expression in cultured cells. The assay uses an indirect ELISA format in which target-specific primary antibodies are detected by HRP-conjugated secondary antibodies, with signal read at 450 nm. It is validated for human, mouse and rat samples and is intended for use with adherent cells. The dynamic range is greater than 5000 cells per well and the assay is performed on a 96-well microplate.

The kit is a convenient, lysate-free, high-throughput option for measuring relative Tuberin/TSC2 levels and for screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density and adjust for plating differences.

Tuberin (also called TSC2) is a tumour suppressor that, together with Hamartin (TSC1), inhibits mTOR nutrient signalling, and its dysregulation is associated with tuberous sclerosis complex. The kit is stored at 4 °C and is stable for 6 months. It is supplied for research use in the field of immunology.
