Description

Designed for the precise measurement of Tryptophan Hydroxylase phosphorylated at serine 58 in cell lysates and tissue samples, this cell-based ELISA kit from Assay Genie offers a high-throughput, lysate-free workflow. The assay is reactive with Human, Mouse, and Rat samples and is recommended for use with adherent cells. Detection is performed by a colorimetric method read at 450 nm, and the kit supports a dynamic range of > 5000 Cells.

The kit uses an indirect ELISA format in which the target is captured by Tryptophan Hydroxylase (Phospho-Ser58)-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. Separate antibodies against the phospho-target and the total target are provided, allowing the relative amounts of phosphorylated Tryptophan Hydroxylase to be measured in cultured cells and the effects of various treatments, inhibitors, or activators on phosphorylation to be screened. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density and adjust for plating differences.

Tryptophan Hydroxylase is the rate-limiting enzyme in the biosynthesis of serotonin, catalysing the biopterin-dependent monooxygenation of tryptophan to 5-hydroxytryptophan (5HT), which is then decarboxylated to form serotonin. Phosphorylation at serine 58 modulates the enzyme's activity, making it a key target for studies in the Neuroscience research area. The target is encoded by the TPH1 gene (Gene ID: 7166; UniProt ID: P17752).
