Description

From Assay Genie, this cell-based ELISA kit is designed for the detection of Thioredoxin Reductase 2 (TRXR2), a selenocysteine-containing mitochondrial flavoenzyme involved in maintaining cellular redox homeostasis and defending against oxidative stress. The assay uses a colorimetric detection method at 450 nm and operates in an indirect ELISA format, in which TRXR2 is captured by specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition.

Validated for human, mouse, and rat samples, the kit provides a lysate-free, high-throughput approach to measuring relative TRXR2 protein expression in cultured adherent cells, and is suitable for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on TRXR2 levels. The dynamic range is greater than 5,000 cells. For data normalization, an anti-GAPDH monoclonal antibody is included as an internal positive control, and a Crystal Violet whole-cell staining method can be used to determine cell density so absorbance values can be adjusted to cell amounts.

TRXR2 is encoded by the TXNRD2 gene (UniProt ID Q9NNW7), which maps to chromosomal location 22q11.21. The protein localizes to the mitochondrial matrix and belongs to the class-I pyridine nucleotide-disulfide oxidoreductase family. Dysfunction of TRXR2 has been implicated in cancer, neurodegenerative diseases, and cardiovascular disorders. The kit is supplied in a microplate format and is stored at 4°C with a stability of 6 months.
