Description

A cell-based ELISA kit from Assay Genie enables qualitative detection of Trk A phosphorylation at tyrosine residues 680 and 681 in cultured cells. Trk A is a receptor tyrosine kinase and a high-affinity receptor for nerve growth factor, neurotrophin-3 and neurotrophin-4/5, and plays a critical role in nerve growth factor signaling pathways.

The kit uses a lysate-free, high-throughput indirect ELISA format in which phosphorylation-specific and target-specific primary antibodies are captured and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition, measured at 450 nm. The assay is reactive with Human, Mouse and Rat samples and has a dynamic range of > 5000 Cells.

A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method can be applied to determine cell density so absorbance values can be normalized to cell amounts. The kit can be used to measure relative amounts of phosphorylated Trk A in cultured cells and to screen the effects of various treatments, inhibitors or activators on Trk A phosphorylation. The target corresponds to the NTRK1 gene (Gene ID: 4914, UniProt ID: P04629), with a molecular weight of 87.511 and two splice variant isoforms described.
