TRIM59 Colorimetric Cell-Based ELISA

Assay Genie's cell-based ELISA kit is designed for the accurate detection of TRIM59 in cell samples, offering a high-sensitivity and high-specificity workflow for research applications. The assay uses a colorimetric detection method read at 450 nm in a 96-well microplate format, with a stated dynamic range of > 5000 cells and reactivity against human and mouse samples. It is intended for use with adherent cells and provides a lysate-free, high-throughput protocol.

TRIM59 is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies then catalyse a colour reaction on substrate addition. Because the readout is qualitative, two normalisation strategies are built in: an anti-GAPDH monoclonal antibody serves as an internal positive control, and Crystal Violet whole-cell staining is used to correct for cell density. The kit can be used to measure relative TRIM59 expression in cultured cells and to screen the effects of treatments, inhibitors such as siRNA, or chemical activators. TRIM59 (tripartite motif-containing protein 59) belongs to the TRIM/RBCC family, has a reported molecular weight of 123,321 Da, and its dysregulation has been linked to diseases including cancer and autoimmune disorders.

Supplied reagents include HRP-conjugated anti-rabbit and anti-mouse IgG antibodies, 10X TBS, blocking and wash buffers, quenching buffer, primary antibody diluent, ready-to-use substrate, stop solution, SDS solution, Crystal Violet solution, anti-phospho and anti-target antibodies, and adhesive plate seals. The kit is stored at 4°C with a stability of 6 months.
