Description

From Assay Genie, this Cell-Based ELISA Kit is designed to detect TRIM3 protein expression directly in cultured cells using a colorimetric readout at 450 nm. It is a lysate-free, high-throughput indirect ELISA format in which TRIM3-specific primary antibodies are bound by HRP-conjugated secondary antibodies, allowing qualitative measurement of relative TRIM3 levels and screening of treatments, inhibitors, or activators in adherent cells.

Reactivity has been validated for Human, Mouse and Rat samples. The assay uses a 96-well microplate format with a dynamic range of greater than 5000 cells per well. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, and Crystal Violet whole-cell staining is provided to determine cell density so that absorbance values can be corrected for plating differences.

TRIM3 (tripartite motif-containing protein 3; UniProt O75382, Gene ID 10612, chromosomal location 11p15.5, molecular weight 80,830 Da) belongs to the TRIM/RBCC family of RING finger proteins and is probably involved in vesicular trafficking via the CART complex, supporting transferrin receptor recycling. It localizes to the cytoplasm and early endosome, exhibits zinc ion binding and ubiquitin-protein ligase activity, and participates in nervous system development and protein ubiquitination. Three isoforms of the human protein are produced by alternative splicing; aliases include BERP, HAC1, RNF22 and RNF97.
