Description

Designed for the qualitative measurement of Transglutaminase 2 protein expression in cultured cells, this cell-based ELISA kit from Assay Genie offers a high-throughput, lysate-free workflow. The assay is reactive with human and mouse samples and is intended for use with adherent cells. Detection is colorimetric, measured at 450 nm, with a dynamic range suitable for samples containing > 5000 cells. The kit enables screening of the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on Transglutaminase 2 levels.

The assay uses an indirect ELISA format in which the target is captured by Transglutaminase 2-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. To account for the qualitative nature of the assay, a monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values. A Crystal Violet whole-cell staining method is also provided for determining cell density, allowing results to be normalized to cell amounts and adjusted for plating differences. The kit is supplied in a 96-well microplate format and is stable for 6 months when stored at 4°C.

Transglutaminase 2 is a calcium-dependent enzyme of the transglutaminase family that catalyzes protein crosslinking and polyamine conjugation. It is involved in cellular processes including apoptosis, inflammation, and tissue remodeling, and its dysregulation has been implicated in cancer, neurodegenerative disorders, and autoimmune conditions. The protein is the autoantigen implicated in coeliac disease.
