Description

From Assay Genie, this cell-based ELISA kit is designed for the quantitative detection of Tryptophan Hydroxylase 2 (TPH2) levels in cell lysates, serum, plasma, and other biological fluids, and is suitable for adherent cells. The assay uses a colorimetric detection method at 450 nm and is reactive with human, mouse, and rat samples.

TPH2 is the rate-limiting enzyme in serotonin biosynthesis, catalysing the biopterin-dependent monooxygenation of tryptophan to 5-hydroxytryptophan. Dysregulation of TPH2 activity has been implicated in psychiatric disorders such as depression, anxiety, and schizophrenia, making this kit relevant to neuroscience research, studies of serotonin signalling, and drug development efforts aimed at modulating serotonin levels in the brain. The kit supports a dynamic range of more than 5000 cells and is supplied in a 96-well microplate format intended for storage at 4°C for up to 6 months.

Described as a convenient, lysate-free and high-throughput assay, the kit detects TPH2 protein expression in cultured cells and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators. TPH2 is captured by TPH2-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyse a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control in normalising the target absorbance values, and a Crystal Violet whole-cell staining step allows cell density to be determined so that absorbance values can be adjusted for plating differences.
