TOP2B Colorimetric Cell-Based ELISA Kit

A Cell-Based ELISA kit from Assay Genie designed for qualitative measurement of TOP2B (DNA topoisomerase 2-beta) protein expression in cultured cells. The assay uses an indirect ELISA format, where TOP2B is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm upon substrate addition. It is reactive with Human and Mouse samples, intended for use with adherent cells, and supplied in a 96-well microplate format with a stated dynamic range of > 5000 Cells. Storage is at 4°C for 6 Months.

The kit supports measurement of relative TOP2B levels in cultured cells, including screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators. Because the readout is qualitative, two normalization strategies are provided: a monoclonal anti-GAPDH antibody serves as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences.

TOP2B encodes a DNA topoisomerase involved in chromosome condensation, chromatid separation, and the relief of torsional stress during DNA transcription and replication. Dysregulation of TOP2B activity has been linked to diseases including cancer and neurological disorders, and the enzyme functions as the target for several anticancer agents.
