TOP2A (Phospho-Ser1525)Colorimetric Cell-Based ELISA Kit

From Assay Genie, this cell-based ELISA kit is designed for the quantitative detection of phosphorylated TOP2A at serine 1525 in cell samples and offers a convenient, lysate-free, high-throughput workflow. It uses an indirect ELISA format in which TOP2A (Phospho-Ser1525)-specific primary antibodies are captured, with HRP-conjugated secondary antibodies catalysing a colorimetric reaction read at 450 nm.

The assay is reactive with Human, Mouse and Rat samples and is intended for adherent cells, with a stated dynamic range of > 5000 Cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining step can be used to determine cell density and correct for plating differences. The kit can be used to measure relative phosphorylated TOP2A levels in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on TOP2A phosphorylation.

TOP2A is a DNA topoisomerase that controls and alters the topological states of DNA during transcription and is essential during mitosis and meiosis for proper segregation of daughter chromosomes. Phosphorylation at serine 1525 has been linked to DNA repair and cell proliferation, and dysregulation of TOP2A activity has been implicated in cancer development and progression, making this phospho-specific cell biology assay relevant to studies of cellular pathways and disease mechanisms.
