TOP2A (Phospho-Ser1106)Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA kit is designed for the accurate detection of phosphorylated TOP2A protein at serine 1106 in cell samples. The kit targets the human TOP2A (Phospho-Ser1106) protein and uses a colorimetric detection method measured at 450 nm. It is a lysate-free, high throughput assay suitable for adherent cell samples, enabling measurement of relative amounts of phosphorylated TOP2A in cultured cells as well as screening for the effects that various treatments, inhibitors, or activators have on TOP2A phosphorylation.

Detection is achieved via an indirect ELISA format in which HRP-conjugated secondary antibodies bind the Fc region of TOP2A (Phospho-Ser1106)-specific primary antibodies, catalysing a colorimetric reaction upon substrate addition. A monoclonal GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density. The dynamic range is > 5000 cells. TOP2A (UniProt P11388, NCBI Gene ID 7153) is a DNA topoisomerase with a molecular weight of 182,681 Da, essential during mitosis and meiosis for proper segregation of daughter chromosomes, and phosphorylation at serine 1106 has been implicated in cancer progression and drug resistance.
