Description

This Cell-Based ELISA from Assay Genie is designed for the accurate detection of TNR16 protein expression profiles in cultured cells, offering a lysate-free, high-throughput and sensitive workflow. It is suited to measuring relative TNR16 levels in cells and to screening the effects of treatments, inhibitors (such as siRNA or chemicals) and activators. The assay uses a 96-well microplate format with a colorimetric detection method at 450 nm, a stated dynamic range of > 5000 cells, and reactivity with human, mouse and rat samples. Storage is at 4°C for 6 months.

Detection follows an indirect ELISA format in which TNR16 is captured by TNR16-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the conjugated HRP to catalyse a colorimetric reaction upon substrate addition. A monoclonal anti-human GAPDH antibody is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining is provided to determine cell density so that absorbance values can be normalised to cell amounts.

The target corresponds to the NGFR gene (Gene ID: 4804; UniProt ID: P08138), also known as CD271, p75NTR or TNFRSF16, a low-affinity nerve growth factor receptor of the tumour necrosis factor receptor superfamily.
