Description

Manufactured by Assay Genie, this Cell-Based ELISA Kit is designed for the qualitative measurement of TNFRSF9 protein expression in cultured cells. The assay uses a colorimetric detection method read at 450 nm on a 96-well microplate format and supports a dynamic range of > 5000 Cells. It is intended for use with human samples and is listed for adherent cells.

The workflow is lysate-free and relies on an indirect ELISA format: TNFRSF9 is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. Because the readout is qualitative, an anti-GAPDH monoclonal antibody is included as an internal positive control for normalisation, and Crystal Violet whole-cell staining is used to determine cell density and adjust for plating differences. The kit can be applied to measure relative TNFRSF9 levels and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators.

TNFRSF9, also known as CD137 or 4-1BB, is a member of the tumour necrosis factor receptor superfamily and functions as a receptor for TNFSF9/4-1BBL. It contributes to the clonal expansion, survival, and development of T cells and is involved in regulating immune responses and inflammation, making it a relevant target for research in autoimmune disease, cancer immunotherapy, and related areas of immune regulation.
