TNFC Colorimetric Cell-Based ELISA

From Assay Genie, this In-Cell ELISA Kit uses a colorimetric detection method to identify TNFC protein expression profiles in cultured cells. The cell-based assay is lysate-free and designed for high throughput, sensitive screening of adherent cell samples.

Reactivity is established for human and mouse samples. Detection is read at 450 nm, with a dynamic range of > 5000 cells. The kit is supplied in a 96-well microplate format and should be stored at 4°C, where it remains stable for 6 months.

Qualitative determination of TNFC concentration is achieved through an indirect ELISA format, in which TNFC-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. An anti-GAPDH monoclonal antibody is included as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining is provided for determining cell density to adjust for plating differences.
