TLE4 Colorimetric Cell-Based ELISA

From Assay Genie, this Cell-Based ELISA Kit offers a convenient, lysate-free, high-throughput and sensitive approach for detecting the TLE4 protein expression profile directly in cultured cells. It is intended for use with adherent cell samples and employs an indirect ELISA format, in which TLE4 is captured by target-specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition.

Because the assay is qualitative, two normalization strategies are built in: a monoclonal anti-GAPDH antibody serves as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences. The kit can be used to measure relative amounts of TLE4 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on TLE4 levels. Detection is colorimetric, measured at 450 nm, with a stated dynamic range of greater than 5000 cells.

TLE4 (Transducin-like enhancer protein 4) is a transcriptional corepressor that plays a role in the regulation of gene expression and cell differentiation, and dysregulation of TLE4 has been linked to cancer and neurodevelopmental disorders.
