Description

A colorimetric cell-based ELISA kit from Assay Genie is designed to detect and quantify TIP60 phosphorylated at Ser86 in cell lysates. The assay shows reactivity with human and mouse samples and is suitable for use with adherent cells. Detection is performed at 450 nm through a colorimetric reaction, with a dynamic range exceeding 5000 cells.

The kit employs an indirect ELISA format in which phosphorylated TIP60 at Ser86 is captured by phospho-specific primary antibodies. HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing the colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density for further data adjustment.

TIP60, encoded by the KAT5 gene, is a histone acetyltransferase of the MYST family originally isolated as an HIV-1 TAT-interactive protein. It plays roles in chromatin remodeling, transcription regulation, DNA repair, and apoptosis. The kit can be used to measure relative amounts of phosphorylated TIP60 in cultured cells and to screen for the effects of various treatments, inhibitors, or activators on TIP60 phosphorylation, within the research area of developmental biology.
