Description

A cell-based ELISA kit from Assay Genie enables detection of Tissue Inhibitor of Metalloproteinase 3 (TIMP3) levels in biological samples. The assay uses a colorimetric detection method at 450 nm in an indirect ELISA format, allowing relative measurement of TIMP3 protein expression in cultured cells. The kit is suitable for adherent cells and is also described for use with serum, plasma, and lysate samples.

TIMP3 regulates the extracellular matrix by inhibiting matrix metalloproteinases (MMPs) involved in tissue remodeling and degradation. Dysregulation of TIMP3 has been associated with cancer, cardiovascular disorders, and inflammatory conditions, making it a relevant biomarker across multiple research fields. Mutations in the TIMP3 gene have also been linked to Sorsby's fundus dystrophy.

The kit reacts with human, mouse, and rat samples and is supplied in a 96-well microplate format, with storage at 4°C and stability over 6 months. A dynamic range of greater than 5000 cells is supported. Within the workflow, TIMP3 is captured by TIMP3-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze the colorimetric reaction upon substrate addition. An anti-GAPDH monoclonal antibody is included as an internal positive control for normalization of target absorbance values. Following colorimetric measurement of HRP activity, a Crystal Violet whole-cell staining step can be used to determine cell density, allowing absorbance values to be normalized to cell amounts.
